System parameters required files
Two types of files—a Profile file and a Signal file—are required for estimating system parameters in MaSTR.
• The required Profile file is the (System Parameters) Single Source Profile file. A Single Source Profile file, also referred to as a genotype file, lists all the single source validation samples and the expected alleles for all the markers that are present in the panel that was used to analyze the single source validation samples. Optionally, the file can also include the signal (peak) height for each allele.
Example Profile file
| Because a Single Source profile is used to establish the parameters for an SOP, it is sometimes referred to as the “expected” profile. |
• The required Signal file is the (System Parameters) Single Source Signal file. A Single Source Signal file is a data file that contains all the samples that were listed in the Single Source Profile file. The samples were analyzed as replicates of single source dilutions according to the lab’s SOPs. The file includes the names of the samples, the panel markers, and the alleles and peak heights for both true peaks and stutter peaks. The file must contain at least one non-OB allele for each marker where OB is a flag that indicates “Out of Bin.”
Example Signal file
Both file types have the following features in common:
• Both file types are tab-delimited text files that have a single header line and one row per marker.
• Both file types must include a sample name in each row of data with the following caveat: If the file stores the information for multiple individuals, then you must give each individual (sample) a unique name.
